Downstream Processing: Purification & Isolation

Using microwave peptide synthesis often results in higher purity crude products, making the downstream purification process easier and more efficient.

Purification scales range from 1g to 40 kg/batch final peptide.

Crude peptides are typically purified by reverse-phase High Pressure Liquid Chromatography (RP-HPLC or HPLC). Using microwave peptide synthesis often results in higher purity crude products, making the purification process easier and more efficient.

Additional purification methods employed by AmbioPharm include crystallization, ion exchange chromatography (IEX), which is particularly useful in the case of pegylated peptides for removal of the free, unreacted PEG, and size exclusion chromatography (SEC) may be used to remove high-molecular-weight impurities and polymers such as with multi-disulfide peptides. AmbioPharm employs a wide range of HPLC columns to handle nearly any scale.

Isolate peptides in a wide variety of scales.

After purification, the resulting peptide is dissolved in rather large volumes of acetic acid and water. It is necessary to obtain the final, purified product in powder form. The techniques employed include lyophilization, precipitation, crystallization, and spray-drying. AmbioPharm has the capacity to isolate purified peptides in a wide variety of scales.
